Read defender (or read key) is the overarching term for any defender whose post-snap movement dictates whether the quarterback hands the ball off, keeps it as a runner, or throws a pass.
Rather than assigning a blocker to every defender in the box, option and RPO schemes intentionally leave one defender unaccounted for in the blocking count. Modern offenses organize read defenders into three specialized roles:
A second-level linebacker (like the Will) or overhang safety on Run-Pass Options (RPOs), read to choose between handing off against a light box or throwing into the pass window vacated behind him.
Leaving the read defender unblocked neutralizes him at the point of attack while preserving offensive blockers to climb and seal the rest of the defense.

A note on how the term is used
Read defender is the broad umbrella term across option and RPO football. Dive keys and pitch keys are option read defenders choosing between two runners, while conflict defenders are RPO read defenders choosing between a run fit and pass coverage.
